We investigate how defects in cellular responses to genotoxic stress contribute to tumourigenesis and how to use them in cancer treatment

+ genome instability, + cell cycle checkpoint, + replication stress, + protein phosphorylation, + cancer, + cancer predisposition

Description of PPM1D/WIP1 truncating mutations

We have identified unusual mutations in the PPM1D gene that express proteins with a truncated C-terminus. These increase the stability of the PPM1D/WIP1 protein while preserving its enzymatic activity. [pubmed] [doi]

Identification of oncogenic properties of truncated PPM1D/WIP1

We have developed a new transgenic mouse model carrying a truncated PPM1D allele and demonstrated its contribution to tumour growth. [pubmed] [doi]

Identification of pathogenic variants of CHEK2 in breast and ovarian cancer

We have developed a new assay for measuring CHK2 kinase activity in cells. In collaboration with experts in medical geneticists, we have classified pathogenic mutations in the CHEK2 gene present in Czech patients with breast and ovarian cancer. [pubmed] [doi]
